چکیده

A ~ t T h e interaction of concanavalin A with a comprehensive series of carbohydrate-bovine serum albumin conjugates was investigated by agar gel diffusion and quantitative precipitation techniques. These studies showed that when conjugated to bovine serum albumin, those sugars which inhibited concanavalin A-polysaccharide interaction formed a precipitate with concanavalin A. The unexpected reactivity of the p-phenylazo /3-D-glucopyranoside-bovine serum albumin conjugate is rationalized on the basis of inhibition data employing several aryl and alkylglycosides.

A Spectrophotometric Study of the Carbohydrate Binding Site of Concanavalin A*

The carbohydrate binding sites of the jack bean lectin, concanavalin A, were studied spectrophotometrically. A number of ligands were examined for their ability to displace the chromogenic ligand p-nitrophenyl cr-n-mannopyranoside. It is shown that this technique provides a simple and convenient approach for obtaining association constants for a series of displacing ligands. The data can best b...

Protein-Carbohydrate Interaction III. Agar Gel-Diffusion Studies on the Interaction of Concanavalin A, a Lectin Isolated from Jack Bean, with Polysaccharidesl

Concanavalin A, a protein isolated from the jack bean (Canavalia ensijormis), has previously been shown to form a precipitate with glycogen and yeast mannan, and more recently with dextrans and amylopectin. Double-diffusion precipitation studies in agar gel have been found to afford a very sensitive and satisfactory method for investigating the interaction of concanavalin A with certain types o...

Carboxyl Groups in Carbohydrate-concanavalin a Interaction

I. The role of carboxyl groups in the binding of specific carbohydrates to concanavalin A, the phytohemagglutinin of the jack bean, has been studied using two techniques: (I) H + titration in the presence and absence of methyl a-D-mannopyranoside, a specifically bound carbohydrate ligand and (2) chemical modification of the protein carboxyl groups employing glycine methyl ester and a water-solu...

Microwave irradiation of whole soybeans in ruminant nutrition: Protein and carbohydrate metabolism in vitro and in situ

Whole soybeans serve as one of the main sources of protein in ruminant nutrition. Different processing methods have been employed for ruminal protein protection. The present study was conducted to determine the effects of microwave irradiation [900 W; 2, 4 and 6 min] on quality, ruminal degradability and estimated in vitro intestinal digestibility of availability soybean crude protein....
